RESUMEN

This study was designed to assess the effect of ginger root extract (GRE) supplementation on the oxidative status and intestinal mucosal development in broiler chickens for 6 weeks. Day-old chicks (Ross 708 strain, n = 432) were distributed into six treatments with six replicate of twelve birds each: Negative CON (basal), MX (basal diet + bacitracin methylene disalicylate (BMD) 0.055 g/kg diet), GRE-1 (basal diet + 0.375% GRE), GRE-2 (basal diet + 0.75% GRE), GRE-3 (basal diet + 1.5% GRE), GRE-4 (basal diet + 3% GRE). Growth indices, goblets cell count, mucin (MUC2) in ileum tissue, antioxidant (SOD, CAT, and GPX) in ileum and liver, biological antioxidant potential (BAP), and reactive oxygen metabolite level in blood and intestinal villi measurement were determined. Body weight (BW) was highest (p < 0.05) in all groups except GRE-4, body weight gain (BWG) was best in GRE-1, while FCR was least in all groups except GRE-4. Optimum MUC2 gene expression, SOD, CAT, blood antioxidants, and intestinal morphometric values were observed in GRE-3. The inclusion of ginger root extract up to 1.5% improved growth and reduced oxidative stress while enhancing mucosal development in broiler chicks.

Herbal extracts have been widely evaluated in poultry production for their beneficial effects and potential substitute for antibiotics, which contribute to AMR and risks to human health through the consumption of infected meat. Salmonellosis is a systemic infection caused by Salmonella, an intracellular bacterium with the ability to cause systemic infections with significant implications for both the health and safety of farmers and consumers. The excessive use of antibiotics has escalated the incidence of antibiotic resistance bacteria in the poultry and livestock industry, highlighting the urgent need for alternatives especially in meat-type poultry. Both in vivo usage and in vitro studies of bioactive compounds from herbal extracts have demonstrated the effective antimicrobial activities against pathogenic bacteria, showing promise in managing Salmonella infections and enhancing poultry performance. Phytobiotic feed additives have shown promising results in improving poultry output due to their pharmacological properties, such as stimulating consumption, and enhancing antioxidant properties and preventing the increasing antimicrobial resistance threats. Despite potential for synergistic effects from plant-derived compounds, a further investigation into is essential to fully understand their role and mechanisms of action, for developing effective delivery systems, and for assessing environmental sustainability in controlling Salmonella in poultry production.

Objective: To establish quality standards for Liuwei Nengxiao pills, to optimize the quality control method, and to provide references for the quality control of Liuwei Nengxiao pills. Methods: Chebula, dried ginger, and Tibetan liqueur root in Liuwei Nengxiao pills of different batch numbers were analyzed by thin layer chromatography (TLC). Then, the content of chrysophanol in the preparation was determined by high performance liquid chromatography (HPLC). Furthermore, a series of methodological validation, including the investigation of the linear relationship, precision, stability, and reproducibility and sample recovery test, were performed to verify the reliability of the results. Results: The TLC identification method was easy to perform and demonstrated high specificity, clear spots, and good separation effect. In addition, the negative controls showed no interference. The HPLC method showed high accuracy. The results of methodological validation showed that the peak area of chrysophanol had a good linear relationship (r2=1.0) in the range of 0.06-0.80 µg, presenting good precision (with the relative standard deviation being lower than 2.0%), good stability and reproducibility (with the relative standard deviation being lower than 1.0%), and an average recovery rate of 100.8%. Conclusion: TLC and HPLC are easy to perform, showing high accuracy and reproducibility. The quality standards established are scientific, reasonable, stable, and feasible, providing references for the quality control of Liuwei Nengxiao pills.

There have been a plethora of studies on the effects of access to runs on chickens' welfare and behavioural repertoire with a paucity of information on the comparative advantage of various legume pasture and deep litter system. A total of 200-day-old unsexed Marshall Broiler chicks were weighed and assigned randomly into five experimental groups, viz. deep litter without access to run (DL), deep litter with access to Stylosanthes hamata (SH), Stylosanthes guanensis (SG), Mucuna pruriens (MP) and free run (FR) during dry season. Each treatment had 4 replicates of 10 birds. Data were collected on growth performance, behaviour, tonic immobility (TI), gait score and blood parameters. The data obtained were subjected to One-Way Analysis of Variance in a Completely Randomized Design. Results showed that the final body weight of birds in SH was significantly higher (p Ë‚ .05) than those of SH, MP, FR and DL which were comparable. The feed intake of the birds of DL, SH and FR was higher than those of the other treatment groups. The feed conversion ratio (FCR) of the birds on legume pastures was lower than those without access to pasture. The birds of SH, SG and MP spent higher (p Ë‚ .05) time drinking, preening, dust-bathing, spot pecking and walking standing behaviours while those in DL and FR spent more (p Ë‚ .05) time feeding. Generally, the gait score of the birds on the different legume pastures was similar but better than those without access to pasture. Tonic immobility of the DL birds was longer than that of FR whose duration was longer than those of the birds on the pastures. The study concluded that access to different legume pastures, particularly Stylosanthes hamata, improved the welfare of broiler chickens without adverse effect on the performance of the birds.

RESUMEN

Maize genomic clones encoding cytokinin-inducible response regulators, ZmRR1 and ZmRR2, have been isolated. In comparison with the corresponding cDNAs, ZmRR2 was found to be interrupted in the translated region by an intron whereas ZmRR1 was not. The 5'-flanking regions of the two genes shared conserved regions and putative cis-elements, which had been identified in maize or other plant species. The run-on transcription assay and the analysis of stable maize transformants of ZmRR1 promoter-beta-glucuronidase fusion gene revealed that the accumulation of the transcripts in response to cytokinins is, at least in parts, attributed by transcriptional activation.

Implication of His-to-Asp and/or Asp-to-His (His-Asp) phosphorelay has been recently reported in signal transduction pathways initiated by ethylene and cytokinin. These signaling systems are generally composed of sensor His-protein kinases, His-containing phosphotransfer (HPt) domains, and response regulator domains. In this study, we isolated maize cDNAs, designated as ZmRR2 and ZmHP2, which encode a response regulator domain and HPt domain, respectively, and we identified their His-to-Asp phosphotransfer activity in vitro. The putative translated product of ZmRR2 was highly similar to that of ZmRR1 (78% identity), a maize response regulator homologue. The putative translated product of ZmHP2 showed similarity to that of HPt domains from Arabidopsis thaliana (AHP1-AHP3: 44 to 47% identity) and Saccharomyces cerevisiae (Ypdlp: 24% identity). In vitro experiments demonstrated that the putative signaling factors can transfer the phosphoryl group from His-80 of ZmHP2 to Asp-90 of ZmRRs. Treating detached leaves with t-zeatin or supplying inorganic nitrogen to the whole plant induced the accumulation of ZmRR1 and ZmRR2 transcripts. On the other hand, the steady-state transcript level of ZmHP2 was not affected by cytokinin or inorganic nitrogen sources. These results indicate that His-Asp phosphotransfer may be involved in the transduction of nitrogen signals mediated by cytokinin, and that multiple response regulators participate in the signaling pathways.

A cDNA clone, pZmCip1, encoding a maize (Zea mays) cytokinin-inducible protein 1 was isolated utilizing the differential display technique, and studied using the expression of ZmCip1 in nitrogen-starved maize plants. The cloned cDNA contained an open reading frame consisting of 157 amino acids with a predicted molecular mass of 16.7 kDa, which possesses similarity with the response-regulators of bacterial two-component signalling systems. In detached leaves, accumulation of ZmCip1 transcript by t-zeatin was dose-dependent in a range of 10(-9) M to 10(-7) M, and occurred within 30 min after treatment. The effect of t-zeatin was replaceable by isopentenyl-adenosine or isopentenyl-adenosine-5'-monophosphate. Pretreatment of detached leaves with cycloheximide did not inhibit the accumulation of the transcript. In whole plants, ZmCip1 transcript was transiently accumulated exclusively in leaves by supply of nitrate or ammonium ions to the roots, whereas the transcript was not accumulated in detached leaves by supply of the nitrogen nutrients. Both the cytokinin- and nitrate-responsive accumulations of ZmCip1 transcript were accompanied by an increase in the immunotitratable protein. Isopentenyladenosine and/or its phosphorylated form(s) accumulated in roots 2 h after supply of nitrate to plants. These results, taken together, suggest that ZmCip1 is a primary response gene to cytokinins, and that it involves, at least in part, the nitrogen-signal transduction mediated by cytokinin in maize.
